I am talking about the left red reflector slightly above the license plate holder. There’s a square hole on the right where another one used to be. Any ideas?

I see you are in the US, so it could be local legislation related.
Normally rear reflectors are incorporated into the rear lamps, but if not, the additional ones are usually larger than those tiny ones you have.
The square holes suggest factory fitted. So, I'm coming back to local legislation demanding them.
I'd be surprised if you ever find another, so you may need to source something from a cycle/motorcycle shop, and find a pair that look acceptable.
 
The square holes are most likely used for holding a large, almost square license plate holder. I have the same holes in my First Series bonnet, when I bought it. Like this:

Side note: in 1976 they changed the license plate sizes (well, at least the rear!) in Italy. Which also explains the slightly different design in bonnet:

As you can see there is a nice, rectangular bit that accommodates the new license plates.

I'm not incredibly familiar with your local regulations, but I've never seen any other lights like that. They seem to small to be rear fog lights and not high enough for third brake lights so I assume it's just because of "styling". Most likely the previous owner wondered what to do with these holes in his bonnet and decided to add reflectors to them to fill in the holes.

Nice to see a 1st series in the US, they're getting quite rare on the continent here and since they were never officially on sale in the US I assume there aren't too many driving around!
