I just want to run this by all in case I am missing the obvious:

Set up
650 126 Engine
Pieburg Electric pump
Fuel Regulator now set to 2 psi

Car just being awoken from winter lay up. Ran OK then no joy. Possible owner contribtion as I removed the mechanical pump (previously left in situ) ran the car wthout a fuel regulator.

Regulator now fitted but Car wont start resulting major flooding of the cylinders.

Logic suggests that the float chambers are stuck open after lay up, but before I pull the carb as there anything else I should consider?

Stuck needle-valve would, logically, be the first thing to check; don't forget the little alloy washer that goes under the needle-valve when you refit it. Also, if you are running with a Weber 26 or 28, check that the 'choke' is fully shutting off. Don't forget, technically it is NOT a choke but a 'fuel enrichment device'--there is no 2nd butterfly to cut off air flow.
2 psi should be fine regarding fuel pressure (but try it a shade lower if you can). I can't find a figure for the standard fuel-pump pressure, but its output should be 5.5galls/25litres per hour.

I would agree with everything that Tom said. Typically small four cylinder engines run at 2.5 psi pump pressure. In addition to checking the float for sticking and correct height setting, also check to make sure it has not developed pin holes and is filling up with fuel causing it to not float. A stuck float can often be freed up by lightly tapping on the carburetor fuel bowl area, although you are probably best to check the carburetor out at this point.

If the cylinders have seen lots of fuel you MUST change the oil.

The Carb is now out for an overhaul but sadly I will be away for a bit before I can refit.

John, a good point re the oil. I used a syringe type pump to remove the petrol but better to be safe than sorrry.

Tin Tin, Not withstanding what Tom has said and the standard output of 25 Lt/Hr I would check that your regulator is doing what it should. The MINIMUM flow a Pierburg Pump will output is 95 Lt/Hr at a static pressure of 0.27 - 0.38 bar the system pressure should be 0.10 bar. (this is for the 7.21440.51.0 pump model) I would also check which pump you actually have as they vary considerably, I have all the details of the range of E1F pumps pm if you need to.
